Separation of Organic Acids & Anions with CElixirOA 5.4
Organic acids and anions play an essential role in both environmental and biological processes. Determination of
these molecules is important for both quality control and research laboratories. This application note shows the
efficiency of CElixirOA to separate anions and organic acids at pH 5.4.
Method Conditions:
Voltage: 30kV
Capillary: MicroSolvCE Bare Fused Silica, 75µm x 60cm
Injection: 5 sec pressure
Run Buffer: CElixirOA pH 5.4 Kit
Detection: Indirect UV 233nm
Polarity: Reverse
Standards Concentration: 20ppm |
Method:
Using a Beckman P/ACE MDQ HPCE System and 75µm ID bare fused silica capillary, the analysis and separation of
over 10 anions and organic acids is easily achieved. The 1.25g of Sugar Molasses was diluted in 100ml of NaOH 0.05M
and was hydrodynamically injected for 5 seconds. The capillary was conditioned according the CElixirOA manual.
The separation was completed in less than 7 minutes. Please note that in reverse polarity, the sample is introduced
at the cathode and migrates to the anode.
Discussion and Rationale:
Many organic acids and small anions are UV transparent, therefore a direct detection method is not usable.
CElixirOA pH 5.4 Kit contains pyridine-dicarboxylic acid (background electrolyte) and a chromophore that
absorbs UV energy at 233nm. Using the dynamic coating technology of CElixir with a polycation and the included
chromophore, the separation shown is possible and easy with no hassles of mixing solutions or preparing any run
buffers on your own.
Comments:
CElixirOA provides simple, reliable and rapid separations covering a wide range of applications. With good
linearity and reproducibility below 0.5% RSD for migration times (click here to view chart) CElixirOA is a
valuable alternative to traditional techniques such as Ion Chromatography.
